Is the Tuolumne Meadows Store in Yosemite open for the season?

Unlikely.
Even though the Tioga Road has been opened, the Tuolumne Meadows campground (and most others outside Yosemite Valley) remain closed.
Keep checking the Yosemite website weekly.
Just announced: Early opening for Reds Meadow:
"Red’s Meadow resort will have its facilities open to the general public starting Memorial Day Weekend! Resupply packages for hikers can be picked up starting Memorial Day weekend."
Vermillion Valley will open May 15.
Muir Trail Ranch will open June 24.
I went over the pass on Tuesday and it was not open. They still need to put the roof on!

I'll bet the store manager got caught off-guard from the early Tioga Pass opening.
