I'm driving from Denver and heading two the rocky mountian national park tomorrow. I'm looking for a good scenic drive that would last 5-6 hours not counting the time driving to and from denver. We would like to be able to see some of the best places in the park in this way: parking at a trailhead and walking a short distance (20-30 mins) OR parking directly at the locaiton. I have two non-hikers with me and don't want to put them through a lot of walking.

Trail Ridge Road is very scenic (if not a bit frightening to this flatlander). You might do a loop through the Park from East-to-West or opposite. Mags will probably chime in with a more-experienced opinion.
If you want scenic then Trail Ridge road up over the top with maybe old fall river road on the way up from the Estes Park side. Pick up a map at the entrance. Lots of short walks possible along the road, especially on the west side, like Lake Irene, Colorado River trail head in to the river, Holzwarth Historic Site. All short walks. Lots of others on the map.
I have an AWD santa fe with a rear differential locker.

Both Trail Ridge Road and Fall River Road are paved with plenty of stopping points along the way. *OLD* Fall River road is dirt. Easy enough to do in your vehicle, but the grade and curves are interesting if you are not used to mountain driving.
The Alpine Visitors Center on Trail Ridge is roughly half way and makes a convenient turnaround point if you don't want to go all the way to Grand Lake.
